Post, Texas

Post is a city in and the county seat of Garza County, Texas, United States. The population was 5,376 at the 2010 census.

History

Post is located on the edge of the caprock escarpment of the Llano Estacado, the southeastern edge of the Great Plains. It is at the crossroads of U.S. Routes 84 and 380.

The land belonged to John Bunyan Slaughter, as it was on his U Lazy S Ranch. In 1906, Slaughter sold it to Charles William (C. W.) Post, the breakfast cereal manufacturer, who founded "Post City" as a utopian colonizing venture in 1907. Post devised the community as a model town. He purchased 200,000 acres (810 km²) of ranchland and established the Double U Company to manage the town's construction. The company built trim houses and numerous structures, which included the Algerita Hotel, a gin, and a textile plant. They planted trees along every street and prohibited alcoholic beverages and brothels. The Double U Company rented and sold farms and houses to settlers. A post office began in a tent during the year of Post City's founding, being established (with the name Post) July 18, 1907, with Frank L. Curtis as first postmaster. Two years later, the town had a school, a bank, and a newspaper, the Post City Post, the same name as the daily in St. Louis, Missouri. The Garza County paper today is called the Post Dispatch. The railroad reached the town in 1910. The town changed its name to "Post" when it incorporated in 1914, the year of C. W. Post's death. By then, Post had a population of 1000, 10 retail businesses, a dentist, a physician, a sanitarium, and Baptist, Methodist, and Presbyterian churches.

Post (Paul Kelly album)

Post is the first solo album by Australian singer-songwriter rock musician, Paul Kelly. Kelly had moved to Sydney by January 1985, after leaving his Melbourne-based Paul Kelly Band and the breakup of his marriage to Hilary Brown.

The album was produced by Clive Shakespeare (Sherbet guitarist) and Kelly, and was released in May 1985 by the independent White Records label, leased to Mushroom Records. The album failed to chart in Australia, with only one single, "From St Kilda to Kings Cross", released in April which also failed to chart. The name of the album, Post relates to both being 'after' significant changes in Kelly's life and to the sense of a 'signpost' to future directions. Kelly dedicated the album to Paul Hewson, keyboardist and songwriter for New Zealand/Australian band Dragon who had died of a heroin overdose in January. Kelly has described Post as a concept album dealing with addictions - not necessarily heroin addiction - but various forms, he has also denied that the songs were autobiographical but that he wrote about the world around him.

Hindi

Hindi (Hindi: हिन्दी hindī), sometimes spelled हिंदी, also called Modern Standard Hindi (Hindi: मानक हिन्दी mānak hindī), is a standardised and Sanskritised register of the Hindustani language. Hindi is the official language of the Union of India.

In the 2001 Indian census, 258 million people in India reported Hindi to be their native language. However, this number includes tens of millions of people who are native speakers of related languages but who consider their speech to be a dialect of Hindi.

Hindi is the fourth most natively spoken language in the world after Mandarin, Spanish and English.

It was envisioned that Hindi would become the sole working language of the Union Government by 1965 (per directives in Article 344 (2) and Article 351), with state governments being free to function in the language of their own choice. However, widespread resistance to the imposition of Hindi on non-native speakers, especially in South India (such as the those in Tamil Nadu), Maharashtra, Andhra Pradesh and West Bengal, led to the passage of the Official Languages Act of 1963, which provided for the continued use of English indefinitely for all official purposes, although the constitutional directive for the Union Government to encourage the spread of Hindi was retained and has strongly influenced its policies.

Plot

The film tells the story of six friends whose mountain trek goes awfully wrong. They encounter bizarre incidents during their mountain trek. These incidents are recorded in a video recorder. HindIII

HindIII (pronounced "Hin D Three") is a type II site-specific deoxyribonuclease restriction enzyme isolated from Haemophilus influenzae that cleaves the DNA palindromic sequence AAGCTT in the presence of the cofactor Mg²⁺ via hydrolysis.

The cleavage of this sequence between the AA's results in 5' overhangs on the DNA called sticky ends:

Restriction endonucleases are used as defense mechanisms in prokaryotic organisms in the restriction modification system. Their primary function is to protect the host genome against invasion by foreign DNA, primarily bacteriophage DNA. There is also evidence that suggests the restriction enzymes may act alongside modification enzymes as selfish elements, or may be involved in genetic recombination and transposition.

Enzyme Structure

The structure of HindIII is complex, and consists of a homodimer. Like other type II restriction endonucleases, it is believed to contain a common structural core comprising four β-sheets and a single α-helix. Each subunit contains 300 amino acids and the predicted molecular mass is 34,950 Da. Despite the importance of this enzyme in molecular biology and DNA technology, little information is available concerning the mechanism of DNA recognition and phosphodiester bond cleavage. However, it is believed that HindIII utilizes a common mechanism of recognition and catalysis of DNA found in other type II enzymes such as EcoRI, BamHI, and BglII. These enzymes contain the amino acid sequence motif PD-(D/E)XK to coordinate Mg²⁺, a cation required to cleave DNA in most type II restriction endonucleases. The cofactor Mg²⁺ is believed to bind water molecules and carry them to the catalytic sites of the enzymes, among other cations. Unlike most documented type II restriction endonucleases, HindIII is unique in that it has little to no catalytic activity when Mg²⁺ is substituted for other cofactors, such as Mn²⁺.
